I have completed Counselling round 1 Choice filling for Nchmct jee. If I am satisfied with Round 1 Choice, Is it still mandatory to fill Round 2?

If you are satisfied with the seat allotment in round 1 of NCHMCT counselling then you need to freeze the seat. It means that you have confirmed the seat and wish to take admission. You won't be eligible to participate in second round if you select freeze option. So
