Admission to IIST is not offered through diploma. Admission to engineering programmes at IIST is offered based on JEE Advanced score. There is minimum qualifying marks for each category, so you have secure at least those qualifying marks/score in JEE Advanced to be eligible for admission to IIST.

Yes, you can take admission in IIST for M.Tech programme.
It takes admission through GATE/JEST/UGC NET.
In order to be eligible you should have complete your B.E/B.Tech/M.Sc or equivalent degree with a minimum 60% marks or 6.5 out of 10 in general category.
